The use of stable isotope labeled tracers and labeled biological samples are well suited to tackle major obstacles of untargeted metabolomics such as the elucidatation of global biochemical composition of biological samples, the study of tracer metabolisation or to improve quantification by internal standardisation. In this study we present novel LC-HRMS based, labeling-assisted workflows and data processing tools (MetExtract and FragExtract) for the untargeted profiling of biological samples and MS/MS spectrum elucidation. The presented approaches involve the measurement of mixtures of 13C labeled and non-labeled biological samples or tracer metabolites. For global metabolomecharacterisation and untargeted profiling of tracer metabolisation, a two dimensional data filtering is performed. First every mass spectrum is inspected for labeling specific MS signal patterns to only extract truly biology derived metabolite ions. Subsequently, MetExtract performs chromatographic peak picking using a wavelet implementation and predicts feature groups to convolute different adducts and in-source fragments of the same metabolite. The resulting data matrix does not only form a reliable basis for metabolite annotation and further statistical analysis but is also excellently suited for evaluating the performance of the complete analytical workflow. A detailed description of the workflow concepts is given and their performance and limitations are discussed. The complete workflows are exemplified with selected biological studies on the characterization of the metabolome of the fungus Fusariumgraminearum and wheat, and the untargeted profiling of mycotoxins in wheat as well as phenylalanine in grapevine. Biography Rainer Schuhmacher is Associate Professor at the University of Natural Resources and Life Sciences, Vienna (BOKU) where he is heading the working group Metabolomics and Bioactive Compounds. Recently, his research focus has shifted from target analytics towards LC-MS and GC-MS based metabolomics of microbes and plants with a special focus on the interaction between living organisms. He received his degrees in Chemistry from the University of Konstanz, Germany and the Vienna University of Technology and in 2009. He earned his habilitation in Analytical Chemistry from the BOKU University. He is (co-)author of more than 80 SCI publications.

This video has been made in the framework of the course “Interdisciplinary excursion on organic agriculture (in Eng.) (EX) - 2018S”, University of Natural Resources and Life Sciences, Vienna (BOKU) . It covered the Bio Alpe-Adria region: South-Eastern part of Austria, North-Western part of Slovenia and North-Eastern part of Italy. The video presents the main challenges that farmers/organizations face in the region. The most influential challenge for some was the lack of knowledge, for others the weather conditions, while others were having troubles related to labor and the ease of machinery use. But despite these challenges they all shared a common vision: to run their farms/enterprises the best way they can in order to feed people with quality food.

The reconstruction of the floodplain inundation is based on water levels and terrain elevations surveyed in 1849. It shows how the floodplain in Vienna was potentially inundated at flows between low water level and a flood with a return frequency of ca. 1 year. Die Rekonstruktion der Überflutung des Augebietes beruht auf Wasserspiegellagen und Geländehöhen, die im Jahr 1849 vermessen wurden. Sie zeigt, wie das Auensystem in Wien bei einer Wasserführung zwischen Niederwasser und einem 1-jährlichen Hochwasser vermutlich überflutet wurde. The reconstruction was made by Mathew Herrnegger as a master thesis in the course of the proVISION-research project "OPTIMA Lobau - Optimised management of riverine landscapes based on a multi-criteria Decision Support System" (project-no. 133-260/05; project leader: Thomas Hein). LWM is a well established international joint degree masters’ programme, fully accredited in Austria, The Netherlands and Kenya. The MSc programme structure/curriculum consists of three semesters of 4-months each, which are held in Austria (University of Natural Resources & Life Sciences, Vienna - BOKU), Kenya (Egerton University) and The Netherlands (IHE Delft), followed by a 6-months research period at one of the three partnering institutions, or your home country (to be decided in consultation with the LWM consortium). Danube river landscape in Vienna 1849 - Flight simulation (Wiener Donau-Auen 1849 - Flugsimulation)

The 3D-reconstruction of the historical river landscape in Vienna is based on a detailed survey of the water bodies and the floodplain terrain in 1849. It shows the situation prior to the major channelization program 1870 - 1875. In the terrain model, the current course of the channelized Danube River is indicated by white lines. The reconstruction was made by Mathew Herrnegger as a master thesis in the course of the proVISION-research project "OPTIMA Lobau - Optimised management of riverine landscapes based on a multi-criteria Decision Support System" (project-no. 133-260/05; project leader: Thomas Hein). The reconstruction of the morphological changes of the river landscape is based on 120 historical maps and surveys. The slides 1715 - 1821 show the fluvial dynamics prior to channelization and those from 1829 - 1925 reflect the situation during and after channelization. The last slide refers to the situation after hydropower plant construction. Die Rekonstruktion der morphologischen Veränderungen der Flusslandschaft basiert auf 120 historischen Karten und Vermessungen. Die Karten 1715 - 1821 zeigen die Flussdynamik vor Beginn der Regulierung, jene von 1829 - 1925 spiegeln die Situation während und nach der Regulierung wieder. Die letzte Karte zeigt den Zustand nach der Errichtung von Donau-Kraftwerken.
